چکیده

Purpose: To investigate the effect of Zhitong Jiangu decoction (ZJD) on osteoarthritis rabbits, and mechanism action involved.
Methods: Chondrocytes were obtained from knees osteoarthritic rabbits. These chondrocytes randomly assigned to 7 groups: sham, 5, 10 20 % normal serum groups; 5 ZJD, ZJD groups. The gross histopathological features rabbit cartilage examined by microscopy. Each group was treated with a different concentration or drug-containing serum. protective concentrations cells determined. Cell proliferation IL-1 MMP-3 determined using cell counting kit (CCK) 8 enzyme-linked immunosorbent assay (ELISA), respectively. mRNA protein expressions inhibitor nuclear factor-κB kinase α (IKK-α) factor kappa B p65 (NF- κB p65) quantitative reverse transcription polymerase chain reaction (qRT- PCR) immunoblotting, as appropriate.
Results: Gross examinations showed that successfully established in knee joint. significantly time- concentration-dependently increased groups, relative sham serum-containing However, NF-κB p65, IL-1, IKK-α markedly reduced groups (p < 0.05).
Conclusion: results indicate mitigates rabbits via regulation signaling pathway. Thus, it can potentially be developed for management osteoarthritis.

Protosappanin A protects against atherosclerosis via anti- hyperlipidemia, anti-inflammation and NF-κB signaling pathway in hyperlipidemic rabbits
Objective(s): Protosappanin A (PrA) is an effective and major ingredient of Caesalpinia sappan L. The current study was aimed to explore the effect of PrA on atherosclerosis (AS). Materials and Methods: Firstly, the experimental model of AS was established in rabbits by two-month feeding of high fat diet. Then, the rabbits were randomly divided into five groups and treated with continuous high ...
متن کاملP133: Targeting NF-Κb Signaling Pathway as Potential Therapeutic with Curcumin in Treatment of Multiple Sclerosis
Curcumin is active component of turmeric and isolated from the rhizome of turmeric, a phenolic natural product. One of inflammatory disease is multiple sclerosis, a multifocal chronic autoimmune inflammatory disease of the CNS, which is also known as a perivascular demyelinating disease. Studies have been shown that neuro-inflammation can have both harmful and beneficial effects on the neuronal...
متن کاملVasoactive intestinal peptide alleviates osteoarthritis effectively via inhibiting NF-κB signaling pathway
BACKGROUND To investigate the treatment effect of vasoactive intestinal peptide (VIP) on osteoarthritis (OA) and the relative mechanism. METHOD The OA model on the SD rat knee was established using the modified Hulth method, and the recombinant pcDNA3.1+/VIP plasmid was constructed. One month after the plasmids VIP were injected intra-articularly into the right knee joint of OA and sham-opera...
متن کاملStaphylococcus Aureus Induces Osteoclastogenesis via the NF-κB Signaling Pathway
BACKGROUND Osteomyelitis is one of the refractory diseases encountered in orthopedics, while Staphylococcus aureus (S. aureus) is the most common causative organism in osteomyelitis. However, the precise mechanisms underlying the bone loss caused by S. aureus infection have not been well defined. Here, we investigated the effect of S. aureus on osteoclast differentiation and the probable molecu...
متن کاملNF-κB Signaling Pathway Confers Neuroblastoma Cells Migration and Invasion Ability via the Regulation of CXCR4
BACKGROUND Accumulating evidence implicates the transcription factor NF-κB as a positive mediator of tumor metastasis, but the molecular mechanism(s) involved in this process remains largely unknown. In this study, we investigated the role of NF-κB signaling pathway in the regulation of CXC chemokine receptor-4 (CXCR4) in neuroblastoma metastasis. MATERIAL AND METHODS NF-κB, CXCR4 mRNA and pr...
